Après avoir obtenu son BA et BS en 1975 à l'Université de Wisconsin-River Falls, Swensen poursuit un doctorat en économie à Yale, où il a écrit sa thèse, « un modèle pour l'évaluation des Corporate Bonds ».
Avant de rejoindre Yale en 1985, le professeur Swensen a passé six ans à Wall Street en tant que Senior Vice Président chez Lehman Brothers, spécialisé dans les activités d'échange, et à titre d'associé en Corporate Finance chez Salomon Brothers, où son travail a porté sur le développement de nouvelles technologies financières.
Il est surtout connu pour avoir inventé ce qui est devenu connu comme "Le modèle de Yale" : une application de la théorie moderne de la gestion du portefeuille boursier.
À la différence des autres fonds universitaires, Yale a massivement réduit ses investissements dans les actions et les obligations américaines pour privilégier des placements dans des fonds moins liquides comme les hedge funds, le private equity ou l'immobilier. Les responsables du fond ne voient pas l'intérêt d'investir dans des actifs moins volatils et plus liquides si les retours sur investissement sont moins élevés.
Le modèle de Yale a été développé par David Swensen et Dean Takahashi, et est décrit de l'ouvrage de Swensen « Innover dans la Gestion de Portefeuille »
David Swensen dirige ce fonds d'investissement, le fameux Yale Endowment, d'une valeur de 17 milliards de dollars. Sur les 10 dernières années, le rendement annuel moyen a été de 17,8% sur ses placements. Swensen a ajouté plus de 16 milliards de dollars dans les caisses de Yale.
Il est l'auteur d'avant-garde de gestion de portefeuille: « An Unconventional Approach to Institutional Investment », qui a été publié par The Free Press en 2000 et « Unconventional Success: A Fundamental Approach to Personal Investment » guide d'investissement pour l'investisseur individuel et qui a été publié par The Free Press en 2005.
En février 2009, Swensen a été nommé par le président Barack Obama, pour un mandat de deux ans au "conseil consultatif pour le redressement économique".
